Mutations in the IDH1 and IDH2 genes are among the most significant genetic alterations in diffuse gliomas and serve as key biomarkers for tumor classification, prognosis, and treatment planning. Studies have shown that IDH1/2 mutations are highly prevalent in lower-grade gliomas (WHO grades II and III) and secondary glioblastomas, where they are associated with better patient survival outcomes. Additionally, these mutations are known to influence tumor metabolism, making them potential targets for emerging therapeutic strategies.
In addition to their role in gliomas, IDH1/2 mutations are found in approximately 20% of acute myeloid leukemia (AML) patients. These mutations disrupt key cellular processes, preventing bone marrow cells from maturing properly and contributing to leukemia development. Targeting these mutations, IDH inhibitors have been developed to help leukemia cells differentiate into normal blood cells. Several of these targeted therapies have been approved for treating AML patients with specific IDH1/2 mutations, highlighting the importance of comprehensive mutation testing for informing therapeutic strategies.
The IDH1/2 Mutation Detection Kit is a real-time PCR-based assay for accurate and sensitive mutation detection. This assay is designed to preferentially amplify mutant DNA, ensuring reliable results even in samples with a high proportion of wild-type DNA.

EntroGenʼs IDH1/2 mutation detection kit requires a real-time PCR instrument capable of detecting FAM and VIC fluorescent probes. This test includes reagents required for PCR amplification/detection, as well as validated reaction controls.
