The MLH1 gene is a key mismatch repair (MMR) gene frequently implicated in Lynch Syndrome (LS), a hereditary condition associated with early-onset colorectal and other cancers. While LS is often caused by germline mutations in MMR genes such as MLH1 and MSH2, gene silencing due to promoter methylation is a common mechanism in sporadic, non-LS tumors exhibiting microsatellite instability (MSI).
The MLH1 Methylation Detection Kit is a real-time PCR-based assay that uses methylation-sensitive restriction enzymes (MSREs) to detect methylation in clinically relevant promoter regions C and D of the MLH1 gene. By comparing fluorescence signals between enzyme-treated and untreated DNA reactions, the assay quantifies methylation levels without the need for bisulfite conversion.
This rapid and cost-efficient assay enables laboratories to screen for MLH1 methylation as a prescreening tool for Lynch Syndrome and to better distinguish between hereditary and sporadic MSI-high tumors.

This test includes reagents required for PCR amplification/detection, as well as validated reaction controls. Each DNA sample is tested in two paired reactions: one is treated with methylation-sensitive restriction enzymes (RE) before qPCR, while the other is left untreated to serve as a reference. The untreated reaction provides a baseline for calculating methylation levels by comparing it to the RE-treated reaction.
Columns and reagents for DNA isolation from plasma are not included. Genomic DNA from FFPE tissues can be extracted using EntroGen pureNA® Genomic DNA Isolation Kit.
