Chromosomal translocations are key genetic events in leukemia, leading to the formation of fusion genes that drive disease progression. One of the most significant translocations in childhood precursor-B-ALL is t(12;21)(p13;q22), resulting in the TEL-AML1 fusion gene, which is present in approximately 25% of childhood precursor-B-ALL cases. This fusion gene plays a crucial role in leukemogenesis by disrupting normal hematopoietic differentiation.
The TEL-AML1 One-Step Detection Kit is a highly sensitive and specific real-time PCR-based assay designed to detect and quantify multiple leukemia-associated fusion transcripts in RNA extracted from blood or bone marrow samples. The assay utilizes a FAM-labeled hydrolysis probe for detecting the target fusion transcripts, along with a Cal Fluor Orange 560 (VIC equivalent)-labeled probe targeting the ABL gene as an internal control. This dual-target design enables accurate quantification using the comparative Ct method (Pfaffl method), eliminating the need for external reference standards.
The use of optimized primers and reagents ensures efficient and reproducible cDNA synthesis in a single-tube reaction, enhancing sensitivity and specificity.
