The ctDNA RAS Mutation Detection Kit is a highly sensitive, real-time PCR-based assay designed for the detection of somatic mutations in KRAS exon 2 and NRAS exons 2 and 3 from circulating tumor DNA (ctDNA) extracted from plasma. Mutations in KRAS (codons 12, 13) and NRAS (codons 12, 13, and 61) are frequently associated with resistance to anti-EGFR therapies in cancers such as metastatic colorectal cancer, highlighting the importance of RAS mutation screening for informing personalized treatment selection.

The assay utilizes allele-specific primers and fluorescent hydrolysis probes to preferentially amplify mutant DNA even in the presence of a high proportion of wild-type DNA. Two reactions detect the following mutations: KRAS G12/G13, NRAS G12/G13 (Reaction A), and NRAS Q61 (Reaction B).

This minimally invasive, plasma-based assay enables detection of clinically relevant RAS mutations when tumor tissue is insufficient or unavailable, offering an important tool for guiding therapeutic decisions in cancer management.
