The DNA Fragmentation Quantification Assay is a qPCR-based assay designed to evaluate the quality and amplifiability of DNA extracted from FFPE and fresh/frozen tissue samples. By amplifying three different fragment sizes (37 bp, 150 bp, and 300 bp) from a reference gene on chromosome 5, the assay provides a comprehensive assessment of DNA integrity.

Each sample is tested against a standard curve, yielding three key outputs:

  • DNA concentration (ng/μl)
  • Amplifiable copy numbers for each fragment size
  • A calculated fragmentation ratio (F-ratio).

These values allow researchers to assess DNA suitability for downstream PCR-based applications and to normalize input amounts more accurately than traditional quantification methods.

 

The DNA Fragmentation Quantification Assay is a simple and efficient qPCR-based assay used to assess the integrity and amplifiability of FFPE-derived DNA. It evaluates fragmentation by amplifying three target sizes—37 bp, 150 bp, and 300 bp—from a reference gene using SYBR® Green detection chemistry. Results are available in under 2 hours.
