Glioblastoma Molecular Markers<\/h3>\n

Glioblastoma multiforme (GBM) represents the deadliest form of brain tumor in humans, with current treatments usually providing only minor benefit in overall survival. Recent advances in understanding molecular subtypes have led to the identification of a number of biomarkers for a more accurate classification of GBM. These molecular markers show potential for better diagnosis and prognosis of GBM patients.<\/p>\n

MGMT promoter methylation status has emerged as an important biomarker for determining responsiveness to\u00a0alkylating chemotherapy .\u00a0 MGMT promoter methylation leads to increased sensitivity to alkylating agents such as temozolomide and has recently been shown to carry strong favorable prognostic significance.<\/p>\n

Kit Features<\/h3>\n

EntroGen’s MGMT promoter methylation kit is a polymerase chain reaction (PCR)-based assay that uses allele-specific primers in a multiplex reaction to identify their target. It works by amplifying the epigenetically silenced MGMT promoter regions after bisulfite treatment of DNA to determine the methylation status of the MGMT promoter.\u00a0 Each kit is supplied with primer\/probe sets for multiplexed detection of both target and internal control genes.\u00a0 A bisulfite conversion kit is also included.<\/p>\n

Testing Procedure and Analysis<\/h3>\n

The testing procedure involves the following simple steps:<\/p>\n

  1. Isolation of DNA or RNA from tumor biopsies, paraffin-embedded sections (FFPE), or fresh frozen tumors.<\/li>\n
  2. Perform bisulfite conversion treatment on extracted DNA.<\/li>\n
  3. Amplification using the provided reagents in the kit.<\/li>\n
  4. Data analysis and interpretation using real-time PCR software.<\/li>\n<\/ol>\n

    EntroGen\u2019s MGMT Methylation Detection Kit requires a real-time PCR instrument capable of detecting FAM and VIC fluorescent probes. The test include reagents required for PCR amplification\/detection, as well as validated reaction controls. Columns and reagents for DNA isolation are not included.\u00a0 Reagents for DNA bisulfite conversion are included.<\/p>\n<\/div>\n
