Chromosomal translocations are among the most significant genetic alterations in leukemia and serve as key biomarkers for disease classification, prognosis, and research into targeted therapies. One of the most well-characterized translocations in leukemia involves the fusion of the BCR gene on chromosome 22 and the ABL gene on chromosome 9, forming the Philadelphia chromosome (Ph). This aberration is particularly common in acute lymphoblastic leukemia (ALL), present in approximately 30% of adult ALL patients and more than 50% of ALL cases in patients over 50 years of age. The resulting fusion gene, BCR-ABL P190 (mbcr), encodes a constitutively active tyrosine kinase that drives uncontrolled cell proliferation and leukemia progression.<\/p>\n

The BCR-ABL P190 (mbcr) One-Step Detection Kit is a highly sensitive and specific real-time PCR-based assay designed to detect and quantify the BCR-ABL P190 fusion transcript in RNA extracted from blood or bone marrow samples. The assay utilizes a FAM-labeled hydrolysis probe for detecting BCR-ABL P190, along with a Cal Fluor Orange 560 (VIC equivalent)-labeled probe targeting the ABL gene as an internal control. This dual-target design enables accurate fusion gene quantification using the comparative Ct method (Pfaffl method), eliminating the need for external reference standards of known copy numbers.<\/p>\n

The use of hot-start DNA polymerase, optimized primers, and hydrolysis probes ensures high specificity, reproducibility, and ultra-sensitive detection, capable of identifying as few as 2 copies of the fusion transcript per 5 \u00b5L of RNA input.<\/p>\n
