JAK2 V617F Mutation

JAK2 is a member of serine-threonine kinases that play a key role in various cellular processes including growth, proliferation, survival and angiogenesis.  JAK2 is a downstream mediator of PI3 kinase.  A somatic mutation that substitutes glutamic acid (E) with lysine (K) at amino acid position 17 results in a constitutively active form of the enzyme that is no longer dependent on activation of upstream components of the pathway.  The JAK2 V617F mutation has been found in various cancers.  Recent studies have shown that tumors harboring the JAK2 V617F mutation are less sensitive to JAK2 inhibitors. Furthermore, tumors with JAK2 V617F mutation are generally found to be negative for EGFR, KRAS and ALK driver mutations.

The JAK2 V617F Mutation Analysis Kit is based on mutation-specific PCR.  Mutation-specific PCR uses primers that are 100% complementary to mutant variants of the gene.  DNA polymerase amplifies only those templates that contain a fully complementary template DNA.  The assay is designed to preferentially amplify mutant DNA in a background of wild-type DNA.

Testing Procedure

The JAK2 V617F Mutation Analysis Kit is a single tube polymerase chain reaction (PCR)-based assay that use allele-specific primers in a multiplex reaction to identify the presence of clinically-actionable V617F mutation.  The assay works by amplifying mutant-specific sequence in samples that contain a mixture of mutant and wild-type DNA and rely on fluorescent probes for detection.

The testing procedure involves three (3) simple steps:

  1. Isolation of DNA from tissue.
  2. Amplification using the provided reagents in the kit.
  3. Data analysis and interpretation.

Equipment and Materials

The JAK2 V617F Mutation Analysis Kit requires a real-time PCR instrument capable of detecting VIC and FAM fluorescent probes simultaneously.

All reagents required for PCR amplification/detection, as well as validated reaction controls are included. Reagents for DNA isolation are sold separately.
